Twilight Sparkle, a unicorn deeply engrossed in her studies, reads about the Elements of Harmony and the prophecy of Nightmare Moon's return on the thousandth year. She frantically writes to Princess Celestia, warning her of the impending danger and the prophecy's fulfillment during the Summer Sun Celebration.
Princess Celestia responds to Twilight's urgent letter, dismissing her concerns about the prophecy as an old legend. Instead, she instructs Twilight to oversee the Summer Sun Celebration preparations in Ponyville and, more importantly, to make friends. Twilight, reluctant and socially awkward, views this as a distraction from her vital research.
Twilight Sparkle begins her task, encountering the eccentric residents of Ponyville: the hospitable Applejack and her large family, the boastful and athletic Rainbow Dash, the fashion-obsessed Rarity, the timid Fluttershy and her animal friends, and the overly enthusiastic Pinkie Pie who throws a surprise party for Twilight. Despite their diverse personalities, Twilight struggles to connect with them, preferring her books.
During the Summer Sun Celebration's eve, Twilight focuses on reading about the Elements of Harmony. At the celebration, Princess Celestia fails to appear. Instead, Nightmare Moon returns, declaring eternal night over Equestria, confirming Twilight's fears and forcing her to confront the reality of the prophecy.
Realizing the prophecy is true and Celestia has disappeared, Twilight understands the urgency of finding the Elements of Harmony. Guided by a book found by Spike, they learn that five of the six elements are in the ancient Castle of the Royal Sisters in the Everfree Forest. Twilight's new acquaintances insist on joining her, emphasizing the importance of sticking together, much to Twilight's initial annoyance.
The group enters the treacherous Everfree Forest. They face several challenges: a collapsing bridge, an injured manticore (which Fluttershy calms by removing a thorn), a confusing, laughing forest that Pinkie Pie navigates with laughter, and a crying sea serpent whose broken mustache Rarity fixes using her own hair, showcasing their unique virtues.
At the castle, Rainbow Dash is tempted by shadowy figures (the Shadowbolts) to join them as their captain, but she chooses to remain with her new friends, demonstrating her loyalty. Twilight finally finds the five elements but realizes the sixth, the Element of Magic, is missing. Nightmare Moon appears, attempting to destroy the elements.
As Nightmare Moon attacks, Twilight identifies each of her friends with an Element of Harmony: Applejack (Honesty), Fluttershy (Kindness), Pinkie Pie (Laughter), Rarity (Generosity), and Rainbow Dash (Loyalty). She realizes that the sixth element, Magic, resides within her and is ignited by the bonds of friendship they share. Together, they use the Elements of Harmony to defeat Nightmare Moon, transforming her back into Princess Luna.
Princess Celestia returns, having given Twilight this mission to discover the magic of friendship. She reconciles with her sister, Luna. Celestia then assigns Twilight a new, ongoing mission: to stay in Ponyville and study the magic of friendship, sending her regular reports. Twilight, now embracing her new friends, happily accepts.
